AN ELECTRON MICROSCOPIC STUDY ON THE RESORPTION AND BONE FORMATION OF DEMINERALIZED BONE MATRIX

Abstract

An electron microscopic study was done to observe the bone formation by
implantation of rabbit demineralized bone matrix into the artificial defect of rabbit
calvaria and the resorption process of the matrix implanted into the rat muscle.
In the calvaria defect, a lot of mesenchymal cells aggregated around the matrix at 1
week. The matrix was resorbed and endochondral ossification was taken place around
the matrix at 2 weeks. Active new bone formation was noted and the matrix was
resorbed at 4 weeks.
At the muscular implantation site, the matrix was partly encapsulated by collagenous
bundle and a lot of mononuclear cells were aggregated. At 2 weeks, multinucleated giant
cells appeared at the surface of the matrix. The giant cells were negative in tartrate
resistant acid phophatase reaction. By light micorscopy, the multinucleated cells
presented the general aspect of osteoclastic cells. At the eletron microscopic observation,
Numberous mitochondria, vacuoles, vesicles and rough endoplasmic reticulum were
dispersed in the cytoplasm of the cell. The cells did not form a ruffled border which is
considered as the typical features of osteoclasts, and projected many cytoplasmic process
into the matrix.
It is suggested that the demineralized bone matrix can induce bone formation by
endochondral ossification and the resorption of the matrix was taken by multinucleated
giant cells and mononuclear cells.
